GAPUZ, SHAIRA P. Analysis of Root Traits Associated with Anaerobic Germination Tolerance in Rice (Oryza sativa L.). Undergraduate Thesis. Bachelor of Science in Biology. Cavite State University, Indang, Cavite. April 2016. Adviser: Dr. Miriam D. Baltazar.
This study aimed to analyze root traits associated with anaerobic germination tolerance in rice (Oryza saliva L.) Specifically, it aimed to: determine rice varieties with AG tolerance in Rice Diversity Panel 1, characterize and identify root traits associated with AG tolerance across rice subpopulation and associate it to possible tolerance mechanisms, and identify polymorphic markers for chromosome 1 of selected genotypes for detection of putative gene for tolerance. Three hundred thirty six varieties from RDP1 were screened for AG tolerance under greenhouse conditions. After 21 days after seeding (DAS), shoot and root length were obtained. Shoot length served as the basis of expression of AG tolerance. The root samples were obtained and subjected for scanning with the aid of WinRhizo Program. Root trait parameters such as root Volume (RV), surface area (SA), and average diameter (AD) were determined and were statistically analyzed using Pearson Correlation Analysis. Twenty four polymorphic markers were screened utilizing eight AG tolerant varieties, IR64 and IR42 as susceptible check. Results revealed that 125 varieties (37%) from RDP1 were found to be tolerant in AG. It include varieties such as Vavilovi, Hatsunishiki, Karabaschak, Sab ini and JC 117. Correlation analysis of several root parameters to AG tolerance showed positive association and this could show tight association with AG tolerance. However, it also revealed a weak correlation of dry weight in AG. In screening polymorphic markers, 19 out of 24 markers expressed polymorphism within the AG tolerance varieties. Three markers were polymorphic between AG tolerance and susceptible check.
Rice Diversity Panel 1 (RDP1) can be a good source of tolerant varieties for further researches and several morphological root characteristics can be tightly associated with AG tolerance. Moreover, polymorphic markers identified were not able to differentiate tolerant to susceptible varieties, thus, these markers cannot be highly recommended for putative gene mapping researches.
|
