ANGELAN, JUVY V. Determination of Hemagglutinating Activity of Lectin From Selected Beetle Species (Coleoptera). Undergraduate Thesis. Bachelor of Science in Agriculture major in Crop Protection. Cavite State University, Indang, Cavite. September 2002. Adviser: Dr. Evelyn 0. Singson.
The study was conducted at the Crop Protection Laboratory, Cavite State University, Indang, Cavite and Biochemistry Research Laboratory, Institute of Chemi stry, University of the Philippines Los Banos (UPLB) College, Laguna from April to May 2002. The main objective of this study was to determine the hemagglutinating activity of lectin from seven beetle species. Lectin extracted from three beetle species exhibited hemagglutinating activity. The three beetle species were: white grub Leuco pholis irrorata), eggplant flea beetle (Psylloides sp.), and the yellow squash beetle (Aulacophora similis). Lectin from other species showed negative hemagglutinating activity. These are corn grain beetle (Sitophilus sp.), coconut click beetle (Rhinocerus pachyrrynchyd beetle (Pachyrrhynchus sp.), and the ground beetle (Rhizopertha Lectin extracted from Leucopholis irrorata had the highest hemagglutinating activity, followed by Psylloides sp. and Aulacophora similis. Characterization of the lectin crude extract from Leucopholis irrorata revealed that it is a non- blood type and non-blood group specific, non-Ca dependent but with higher activity under 30°C than at 4°C. Addition of sugars had no effect on the hemagglutinating activity of lectin.
