Abstract: CUADRA, ALYSSA JANE E. Screening of Poly-B-hydroxybutyrate (PHB) - synthesizing Bacteria Isolated from Various Samples. Undergraduate Thesis, Bachelor of Science in Biology, Major in Microbiology, Cavite State University, Indang, Cavite. June 2019. Adviser: Dr. Yolanda A. Ilagan This study was conducted to isolate, screen, and identify bacterial isolates capable of producing poly-B-hydroxybutyrate (PHB) from various samples. It was conducted at the Microbiology Laboratory of the Research Center, Cavite State University, Indang Cavite from September 2018 to March 2019. Two hundred sixty-eight bacterial isolates were screened for possible PHB production using Sudan Black B plate assay. Results revealed that 20 bacterial isolates were found positive based on high color intensity, and detection of intracellular lipid inclusions by Sudan Black B Staining. The PHB producing isolates were characterized culturally, morphologically and physiologically to determine their identities at generic level. Following the scheme of identification, the isolates were presumptively identified under the genus Bacillus.
