Hagan The study was conducted at Biotechnology Laboratory of the Biological Science Department from October 2006 to January 2007. This study aimed develop a plant DNA isolation protocol suitable for both softwood and hardwood plant species. The used protocols were based on Edwards et al. (1991) and Doyle and Doyle (1990) protocoL The key steps for the modified Edwards et al. (1991) protocol were: 1) extraction with isolation buffer containing SDS (sodium dodecyl sulfate); and 2) precipitation of DNA by isopropanol. Modifications employed on different plant samples were the change in the incubation temperature on both key steps. Electrophoretic analysis showed that it is unsuccessful in extracting DNA in both pechay and coffee. The key steps for the modified Doyle and Doyle (1990) protocol were: (1) extraction with high salt CTAB buffer; (2) chloroform isoamyl alcohol extraction; (3) isopropyl alcohol precipitation, and; (4) cleaning and washing with ethanol. Modifications employed on different plant samples were the change in incubation temperature for CTAB buffer extraction and isopropanol precipitation. Electrophoretic analysis showed fire type and smeared bands on all modifications. The modified Edwards et aL (1991) and Doyle and Doyle (1990) DNA extraction protocol were not successful in producing good quality DNA solution.
