Adviser: Dr. Yolanda A. llagan This study was conducted at the Microbiological Research Laboratory from June 1997 to February 1998 to determine what local substrate and what particular composition could be developed to obtain locally formulated media to be used in supporting the growth of bacteria and fungi comparable to the dehydrated media. Each local substitute was composed of five (5) treatments and two (2) replicates for each of the two trials. This was arranged in Complete Randomized Design (CRD). Results revealed that there was no significant among the substrates used. Thus, locally available substrates which included cassava, sweet potato, rice washing, aerial potato, brown sugar, refined sugar, washed sugar, soymilk, mongo extract, coconut, water and local agar bar or gelatin could be used as effective culture media for growth of bacteria and fungi. Analysis of variance also revealed that there was no significant difference on the control media and the resulting PDA and TGYA based media.
