Baltazar, PhD. <br/>The study was conducted from November 2014 to February 2015 at the Coffee Tissue Culture Laboratory of the National Coffee Research and Development Center, Cavite State University, Indang, Cavite, to identify the effects of hormone pre-treatments and varying photoperiods in the somatic embryo (SE) induction of coffee C. arabica variety Typica. Specifically, the study aimed to determine the following: effects of the duration of hormone pm-treatments; effects of different photoperiods; percent of SE induction in each treatment combination and the best pre-treatment duration and photoperiod in the induction of SE. <br/>The leaves ('. arabica var. Typica were sterilized, then cut to 1cm2 and soaked in 1 ppm NAA and 0.5 ppm kinetin solution for two different pm-treatment durations: 12 hours and 24 hours. Another set of explants remained untreated to serve as control without hormone pre-treatment). The pre-treated explants were cultured abaxial side up in a direct embryogenesis induction medium. The inoculated explants were subjected to complete darkness, 24 hours continuous light, 12 hours/day-' and 16 hours/day-1 2hotoperiods. SE induction was observed directly from the edges of the explant. <br/>At Day 60, best treatment combination of hormone pre-treatment under a photoperiod was observed. Hormone pre-treatment of 12 hours under complete darkness showed the highest percentage of somatic embryogenesis, while the lowest and the slowest response was obtained in the pre-treatment of 24 hours under complete light photoperiod. In terms of the duration of pre-treatment, 12 hours got the highest induction of SE, followed by the control and 24 hours. In terms of different photoperiods, complete darkness accounts for the highest induction, followed by 16/8 hours (Light/dark), 12/12 hours (Light/dark) and continuous light. The study revealed that soaking the explants at 0.1 ppm NAA and 0.5 ppm kinetin solution for 12 hours subjected to complete darkness was the best in the SE induction of C. arabica var. Tvpica. The SE induction of coffee was affected by light and pre-treatment with plant growth hormone, both acting as individual factors. The time response for SE induction was enhanced by the pre-treatment of 12 hours. Complete darkness was the photoperiod suited for the SE induction. This can enhance the next stage of somatic embryogenesis of coffee, since this pre-treatment plus photoperiod are very vital, thus, can be adopted to optimize the SE induction of Carabica.<br/> |
