College of Arts and Sciences, Cavite State University, Indang, Cavite. May 2017. Adviser: Dr. Hosea L. Matel<br/>This study aimed to isolate and identify lactase-producing bacteria from different<br/>raw milk and characterize the produced lactase. In this study, bacteria were isolated from cow, carabao, and goat raw milk obtained from Dasmarifias and Indang, Cavite.<br/>Fifteen isolates were obtained from raw milk using MacConkey agar. A total of 4 isolates were lactase-producer based on lactase assay. Most of the lactase activity on both<br/>supernatant and cell reached the highest point at 48 hours and down at 72 hours. Isolate 5COB2 (obtained from cow) produced highest lactase activity among the isolates at 48<br/>hours at room temperature, also much higher compared to those observed at 370Celsius.<br/>Isolate 5COB2 might produce extracellular lactase due to the observed high lactase activity in the supernatant..Morphological and physiological characteristics of the isolates were studied to identify the organisms. All the four isolates were identified as Klebsiella oxytoca based on the following characteristics; gram negative (-) reaction, rod shaped, positive in VP test, citrate utilization and indole, and negative in hydrogen sulfide production<br/> |
