Baltazar.<br/><br/>The study was conducted at the Tissue Culture Laboratory, National Coffee Research Development and Extension Center (NCRDEC), Cavite State University-Main Campus from September 2015 to January 2016. Generally, it aimed to determine the effect of photoperiod and pre-treatment on induction of somatic embryogenesis in coffee (Coffea arabica var. ‘Red Bourbon). Specifically, this study aimed to: 1) determine the percent of somaiic embryogenesis induction of each treatment combination, 2) determine the number of days for the somatic embryos to appear, 3) determine the effect of photoperiod and PGR pre-treatment on the induction of somatic embryos in terms of: a) percent of somatic embryo induction, and b) number of days to somatic embryo appearance; and 4) determine the color of the somatic embryos induced.<br/><br/>The first treatment to develop SE was LP, at 11 DAC. LyPo and LoP, followed at 14 DAC. The fastest SE response was observed at continuous light (L1) and PGR pre-<br/>treatment. The developed regenerating calli — like tissues started from yellowish white color, turning yellow, then brown to dark brown. White - colored calli — like tissues developed from the dark brown tissues. Photoperiod, pre-treatment, and the combination of these two factors affected the results in terms of percent of SE induction and days to somatic embryo appearance. LoPo, a combination of total darkness and without pre-treatment, is the best treatment in terms of both days to somatic embryo appearance and percent of SE induction.<br/><br/> |
