Sangalang, RMT This study aimed to determine the ability of Bunga (Areca catechu) extract in staining the cyst of Entamoeba histolytica and Entamoeba coli based on, pH level, nuclear and cytoplasmic detail, color stability and staining quality. The main purpose of the study is to determine the staining ability of Areca catechu (Bunga) extract in diagnosing the cyst of Entamoeba histolytica and Entamoeba coli against Lugol's iodine in wet mount preparation. Four treatments and a control replicated three times were used in the study, in five samples here allocated to each treatment. In three days the stained specimen was observe for its color stability and reagent strips was used for pH level identification. The researchers made a description to categorize the staining ability of every treatment used in the study. The study shows that Areca catechu extract used as an alternative stain to Lugol's iodine exhibit poor color stability, staining quality, nuclear and cytoplasmic detail. And it has poor staining ability against the cyst of Entamoeba histolytica and Entamoeba coli. All concentration of Bunga (Areca catechu) extracts was highly significant compared to the staining ability of Lugol's Iodine. The both control and treatment exhibit slightly acidic pH, In wet mount preparation Lugol 's iodine stained clearly to the cyst of Entamoeba hystolitica and coli in which the Bunga extract failed to demonstrate.
