Abstract: SAGPAO, ROY ALESSANDRO N. April 2007. Cavite State University, Indang, Cavite. Isolation, Characterization and Antibiotic Sensitivity Testing of Hemolysin — Producing Escherichia coil 0157:H7 in Carrier Dairy Cattle of the Dairy Training and Research Institute (DTRI) Laguna, Philippines. Adviser: Ma. Cynthia N. Rundina - dela Cruz, DVM, MS. The aims of this study were to isolate and characterize enterohemorrhagic Escherichia coli (EHEC) isolates in rectal swabs of five healthy cattle previously identified as harbouring Escherichia coil 0157:H7, determine the occurrence of hemolysin-producing E. coil 0157:H7, and perform antibiotic sensitivity testing of isolates. against selected antimicrobials using the disk diffusion method. Eighty-three colonies were isolated from Sorbitol MacConkey Agar (Acumedia®) supplemented with cefixime and tellurite (Dynal®). The isolates were further characterized by biochemical and serological 'means and five isolates were identified as E. coil 0157:non H7 and two were E. coil 0157:H7. Moreover hemolysin was detected in three isolates using washed sheep red blood cells wherein one belonged to the 0157:H7 serotype. All isolates were found to be susceptible to the following antibiotics: doxycycline, norfloxacin, colistin, amoxicillin, streptomycin, ciprofloxacin, trimethoprim-sulfisoxazole, and fosfomycin. Whereas, one isolate has intermediate susceptibility to streptomycin. Three of the five dairy cattle were found to have persistent carriage of E. coil 0157 indicating that the organism could be maintained in these animals and thus, serve as potential sources of the organism to other animals and humans.
