Abstract: BANIQUED, APRILEEN T., Applied Research IV (General Science Curriculum) Cavite State University, Indang, Cavite, March 1999. “ In Vitro Germination of Makapuno Embryo Using Solid and Semi-solid Culture Media”. Adviser: Prof Eleanor G. Ersando. This research study entitled “ In Vitro Germination of Makapuno Embryo Using Solid and Semi-solid Culture Media” was conducted at the Makapuno Embryo Culture Laboratory in Crop Science Department of the Cavite State University, Indang, Cavite from April to August 1998. It aimed to: (1) determine the quality of solid culture medium (liquid Y3 medium with 109/L of agar-agar) that will initiate early shoot and root formation on makapuno embryos; and (2) identify the quality of semi-solid culture media (liquid Y3 with 59/L of agar-agar) that will give early germination of makapuno embryo. A total of sixty (60) makapuno embryos purchased from the farmers of San Pablo, Laguna were arranged in Completely Randomized Design ( CRD ). The treatments used in the study were: To — Liquid Y3 culture media, Tl - Solid culture media (liquid Y3 medium with 10 g/L of agar-agar) and T2 — Semi-solid culture media (liquid Y3 medium with 5 p/L of agar-agar). The study revealed that To or the treatment control was the best treatment in terms of root length, growth increment, number of root hairs and number of shoots. On the other hand, T2 or the semi-solid culture media revealed to be the poorest.
