FERRAER, DESTINE MARCEL P., IJAY N. MONTEMAYOR Isolation and Characterization of Cyanobacteria for Hydrocarbon Production.Research Study.Science High School, College of Education, Cavite State University, Indang, Cavite. April 2015. Adviser: Dr. Hosea dL. Matel.
This study was conducted at the Research Center of the Cavite State University. It aimed to determine the species of the Cyanobacteria that will produce hydrocarbons; determine the morphological characteristics of the Cyanobacteria that produce hydrocarbons in terms of color, shape, form, elevation, margin, optical property, and pigmentation; and determine the rate of multiplication of the Cyanobacteria. The water samples were gathered from 9 different sites of CvSU-Naic Campus shoreline. The water samples were transferred to the petri dishes and the bacteria that grew were isolated and cultured in test tube slants. Nine test tube slants with bacteria were chosen as principal samples based on purity. The morphology of the bacteria in the test tube slants were observed under a microscope and by the naked eye. The 9 principal samples were also cultured in sterilized BG-1 I broth. The rates of multiplication were determined by monitoring the absorbance of the samples for 2 weeks. The samples in the broth were also observed under a microscope and compared to the atlas of Laboratory of Phytoplankton Ecology to see if it is Cyanobacteria. The confirmed Cyanobacteria were observed under fluorescence microscope to determine if it produced hydrocarbon. Based on the results of the study, 6 strains of Cyanobacteria were isolated from 9 different sites of CvSU-Naic Campus shoreline. The 6 confirmed samples were Tabellaria flocculosa, Anabaena macrospora, Aphanothece granulosa, Closterium moniliferum, Synechoccus sp. and Cosmarium bot-rytis. All of the said samples produced hydrocarbons.
The rates of multiplication of the samples were 0.020 absorbance unit per hour for sample with Aphanothece granulosa, 0.006 absorbance unit per hour for sample with sample Cosmarium botrytis, 0.018 absorbance unit per hour for sample with Tabellaria flocculosa, 0.013 absorbance unit per hour for sample with Closterium monilijerum, 0.009 absorbance unit per hour for sample with Synechoccus sp., and 0.019 absorbance unit per hour for sample with Anabaena macrospora. The color, shape, form, elevation, margin, optical property and pigmentation of Tabellaria flocculosa sample were colorless and green, square-like, irregular, flat, entire, translucent, and non-pigmented respectively; for Anabaena macrospora sample were blackish, bacillus, irregular, umbonate, entire, translucent, and non-pigmented respectively; for Aphanothece granulose sample were colorless, coccus, irregular, umbonate, entire, opaque, and pigmented respectively; for Closterium moniliferum sample were green, bacillus, irregular, umbonate, entire, opaque, and pigmented respectively; for Synechoccus sp. sample were dark green, bacillus, irregular, umbonate, entire, opaque, and pigmented respectively; for Cosmarium botrytis sample were dark green, coccus, irregular, flat, no margin, translucent, and pigmented respectively.
