Abstract: BABELONIA, JONATHAN A.,PCR Detection of Mt-rrs Gene and Rickettsia spp. among Ticks Collected from Cattle and Pasture in Cavite. Undergraduate Thesis. Doctor of Veterinary Medicine. Cavite State University, Indang, Cavite,2017. Adviser: Dr Emmanuel Mago A study was conducted to have a preliminary survey of the occurrence of Rickettsia pathogens present in Cavite using molecular detection. Tick borne pathogen is of human and animal health concern. Determination of the detection rate of Rickettsia sp. is one of the main objectives of the study. Two hundred sixty-four 264 ticks were collected during the months of April and May. Ticks collected were classified according to species, sex and development stage. DNA samples were extracted using two methods boiling and black Prep Kit. Samples extracted using the kit showed higher detection rate of meters gene. One hundred sixty-five (165) DNA samples were detected having a meter’s gene screening and proceeded for the detection of pathogens. Detection of rickettsia pathogens were done via amplification of citrate synthase (gltA) gene using a nested PCR. All tick samples showed negative results in the test conducted. Results showed that no distinct bands were present in the target 500 bp. It is highly suggestive that cattle farms from Cavite has zero to very low detection rate of rickettsia pathogens .
