EFFECT OF STORAGE DURATION ON THE GERMINATION OF MACAPUNO EMBRYOS IN- VITRO. Prof. Alejandro C. Mojica and Dr. Julio G. Alava, advisers. A study was conducted to determine the effect of storage duration of macapuno nuts on the germination of macapuno embryos in-vitro. The study was laid out in a Completely Randomized Design with four treatments and three replications. The treatments used were: Control (C) - embryos were cultured from macapuno nuts immediately after harvest; Treatment 1 (T1) - embryos were cultured from macapuno nuts one day after harvest ; Treatment 2 (T2) - embryos were cultured from macapuno nuts three days after harvest, and Treatment 3 ([3) - embryos were cultured from macapuno nuts five days after harvest. The study revealed that Treatment 3 significantly increased the percentage germination of macapuno embryos in-vitro and also had the highest number and length (mm) of roots . The average shoot length of developed macapuno plant in-vitro was also observed from two weeks after planting (2 WAP) to twelve weeks after planting (12 WAP). No significant result was observed from 2 WAP up to 4 WAP. However , Treatment 3 ( embryo stored for five days) remarkably increased the shoot length (mm) of developed macapuno plants in-vitro starting from six weeks after planting and the rest of the growth stages of the macapuno seedlings.
