SAPUTIL, KYRIA H. Antifungal Spectrum of Fungal Endophytes Isolated from Young and Matured Leaves of Coffea spp. in Selected Towns of Cavite. Undergraduate Thesis. Taglinao
This study was conducted to isolate and identify fungal endophytes found in young and matured healthy leaves of Coffea spp. positive for inhibition against yeast and molds. Tissue planting method was used to isolate endophytic fungi using Potato Dextrose Agar (PDA) medium amended with chloramphenicol and incubated for 7-14 days. A total of 40 fungal endophytes were isolated from young and matured leaves of Coffea spp. from coffee growing towns of Cavite, namely: Mendez, Indang, Amadeo, Silang and Alfonso. Out of these isolate fungal endophytes, 13 were obtained from the young leaves while 27 were obtained from the matured leaves. These fungal endophytes were screened for their antifungal activity against yeasts such as, Saccharomyces cerevisiae, Candida albicans and Kloeckera, and molds, Aspergillus brasiliensis, Penicillum sp., Fusarium oxysporum and Trichophyton mentagrophyte using agar plug method. One isolate from midrib of matured leaves was positive for inhibition against the A. brasiliensis and T. mentagrophytes. Fungal endophyte that has antifungal activity was identified into the genus, Geotrichum. sp.
