Adviser: Dr. Yolanda A. llagan This study was conducted to identify lactic acid bacteria isolated from selected fermented fruits and vegetables through 16S rRNA gene sequencing. It was conducted at the Microbiology Laboratory of the Research Center, Cavite State University, Indang Cavite from July 2022 to December 2022. Twenty-nine presumptive LAB were isolated from selected fermented fruits and vegetables. Twenty selected LAB isolates were then subjected to phenotypic characterization and were classified according to their presumptive genera. The twenty selected isolates were submitted for gene sequencing but only twelve were identified using 16S rRNA gene sequence analysis. Phylogenetic analysis was performed to investigate the relationship between the LAB isolates and reference strains from different fermented foods in Asia. Based on their phenotypic characteristics, the LAB isolates were classified as Enterococcus (2 isolates), Pediococcus (2), and Lactobacillus (16). 16S rRNA gene sequence analysis identified 8 isolates on species level, namely: Lactobacillus plantarum (4 isolates), Pediococcus pentosaceus (2), Enterococcus hirae, and Enterococcus faecalis. Four isolates were identified as Lactobacillus spp. due to the low quality of their nucleotide sequences. Phylogenetic analysis showed close relationships of 7 isolated LAB with reference strains isolated from different fermented foods which have been investigated for probiotic potential.
